Design Notes

pAH57 was designed by not designed by our iGEM team but by Haldimann and Wanner (2001). This plasmid is the Lambda excisionase helper plasmid and is necessary for the excision of BBa_K617000 from the Lambda attB site of an E. coli chromosome.

The design is analogous to that of K617001 (pINT-ts) except for the addition of Lambda Xis ORF upstream of the Lambda Int ORF.

This plasmid is temperature sensitive and must be propogated at 30 degrees Celsius, incubation at 37 degrees will cure the plasmid.
